Superinfection exclusion by bacteriophage T7.
Only two of the early genes of bacteriophage T7 were found to play a significant role in exclusion of superinfecting bacteriophage T3 particles; genes 0.3 and 1. Protein synthesis by the preinfecting phage particle was not required for efficient exclusion. These findings are discussed with regard to the known functions of these genes during T7 development.

Natural biological systems are selected by evolution to continue to exist and evolve. Evolution likely gives rise to complicated systems that are difficult to understand and manipulate. Here, we redesign the genome of a natural biological system, bacteriophage T7, in order to specify an engineered surrogate that, if viable, would be easier to study and extend. Our initial design goals were to p...
متن کاملNegative control of protein synthesis after infection with bacteriophage T7.
T7 phage induces two negative control mechanisms of protein synthesis: (a) Host-gene expression is repressed by a "T7 repressor," and (b) early T7 protein synthesis is inhibited by a late phage protein.(a) The repressor for host enzyme synthesis is an early T7 protein. Its gene is none of the known early genes; it is located promotor-proximal to gene 1. The repressor function of this protein ca...
متن کاملMethylmercury-induced sedimentation heterogeneity of T7 bacteriophage DNA.
Single-stranded and methylmercurated T7 DNA is composed of two species as evidenced from the sedimentation pattern displayed during band-sedimentation in self-generating density gradients as well as during equilibrium-sedimentation in CS2SO4 density gradients. Under the given experimental conditions, viz., at pH 9.18 and in presence of 0.1 mM CH3HgOH, the two species band in CS2SO4 with a densi...
متن کاملFlap endonuclease of bacteriophage T7
Gene 6 protein of bacteriophage T7 has 5'-3'-exonuclease activity specific for duplex DNA. We have found that gene 6 protein also has flap endonuclease activity. The flap endonuclease activity is considerably weaker than the exonuclease activity. Unlike the human homolog of gene 6 protein, the flap endonuclease activity of gene 6 protein is dependent on the length of the 5'-flap. This dependenc...
